Inhibitory Effects of Mori Cortex on Compound 48/80-induced Histamine Release and Calcium Uptake of Rat Peritoneal Mast Cells

Abstract
Mori Cortex(Morus alba L.), the root bark of mulberry tree has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. Previous studies have demonstrated that Mori Cortex had inhibitory effects on compound 48/80-induced
degranulation and histamine release from rat pertitoneal mast cells9RPMC). The purpose of this study is to determine whether Mori Cortex could inhibit the compound 48/80-inducedd calcium uptake of RPMC during degranulation and histamine release.
Various doses of compound 48/80(0.1, 0.25, 0.5, 1.0¥ìg/ml) were added into the RPMC suspension(10E6 cells/ml) pretreated with or without various doses of Mori Cortex(6.491¡¿10E3, 6.491¡¿1-E2, 6.491¡¿10, 6.491¥ìg/ml) at 37¡Éfor 10 mininutes. Ten minutes later, histamine in the assay and radioisotope calcium uptake method.
Results are 1) the compound 48/80-induced degranulation and histamine release of control without Mori Cortex pretreatment, 2) the elevated calcium uptake of RPMC was induced by increased concentration of compound 48/80 with dose dependent fashion, and 3) Mori Corte inhibits the compound 48/80-induced calcium uptake of RPMC remarkably.
From thed above results, it is suggested that Mori Cortex contains some substances to inhbit the compound 48/80-induced degranulation, histamine release and increased calcium uptake of RPMC.
